Do i have to fish for them where there is a sparkling spot or anywhere will do?

Is clam from fishing too?

Anywhere works. I've only gotten bug fragments from sparkling spots.

For the clam and greenling, you need to fish in the second biome.

I've gotten random hides and crystals from sparkly spots. I think greenling and clam have to be reliably caught while fishing in the endless forest biome.
