I'm struggling this this one.
Which Mutators do you recommend?

The 3 mutators in Quest E will do, they give +116% score. You get ~6k for winning, then you need to save up to 200+ gold after the last night. And you need the no-restart 10% bonus.

I finally got my 26th achievement by beating is stage on a only buildings build with Turtle God, God of Death, and No unit Mutators. 135% bonus score

what weapon / perks did you use?

I just completed this quest 5 minutes ago
My weapon was LightningStaff, my perks were HeavyArmor, TreasureHunter, AncientShrines, my mutators were ChallengeTheTurtleGod, ChallengeTheWaspGod, ChallengeTheGodOfDeath, NoUnitsPact, and my score was 21K.

But with 5 perks instead of just 3, this quest looks like walk in the park :)
I've added to my above-described loadout BigHarbors and CastleBlueprints perks, and easily got 24K as my final score
