For your first question: Unfortunately, you cannot change the keyboard layout for Player One. To play in Co-Op mode, both players need to use controllers. This is a common setup for many Steam games to ensure a consistent and enjoyable gaming experience.
For your second question: To play My Little Pony: A Zephyr Heights Mystery in Co-Op mode, you need to complete the tutorial first (the first mission). Once you finish it, a tutorial on how to play in Co-Op mode will be triggered. Essentially, you just need to press the start button on the second controller, and the second player will join the game. Please note that Co-Op mode will only be available after completing the first mission.
