2) Speechcraft is a tedious skill to level. I usually don't bother with it intentionally. I just use it when I need to. I recommend leaving it as a minor skill and just use it as often as you want.
3) I can't really say what is best for an Assassin Build since I have never tried to make an Assassin, so maybe someone else can help you with that.
I will list a build for you to try if you want incase no one else replies, but I wouldn't say it is the best for Assassin just something you might be able to use if you can't decide.
Race: Your choice
Birthsign: Thief or Warrior
Specialization: Stealth
Favored Attributes: Your choice. I prefer Endurance and Luck or Strength.
Major Skills
Alchemy
Blade
Conjuration
Light or Heavy Armor. Whichever you plan to use.
Marksman
Mercantile
Sneak
I really don't recommend using security or acrobatics as major skills, which are both included in the default Assassin Class.
Race: The best assassins at the start of a game are Dark Elves, Khajiit or Wood Elves, but in the end it doesn't make that much of a difference.
Birthsign: Thief helps stealth generally, Warrior helps when fighting in melee (something assassins may need to do), you may also want to try the Shadow, Invisibility once per day, not that strong but it could help if you find yourself in over your head (you might), use the Shadow to run away if you need to, don't rely on it to see you through the game once a day isn't much.
Favored Attributes: Assassins gain the most from Strength or Agility, but like the race selection it doesn't do very much in the end, so pick what you like.
Skills:
Alchemy- use for poisons to make your first strike as deadly as possible, and potions in case your enemy doesn't go down quickly, potions can help you survive.
Blade- use a dagger or a short sword, the smaller the weapon the better the sneak attack multiplier, you might want to go hand-to-hand if you are using a Khajiit.
Conjuration- can help deal with opponents that don't go down, generally enemies will attack the creature you summon instead of attacking you, it also lets you start with the bound Dagger spell, the summoned creatures fail to measure up to most of the late game enemies and are largely a waste of Magicka past level 20. You could replace Conjuration with Illusion, if you paralyze an opponent who hasn't seen you your sneak attack works until the spell wears off, with a good paralysis spell that is 5 or more sneak attacks in a row. Invisibility is a great deffensive spell, when you cast it your enemies will not be able to find you unless they have an item that lets them "See Life", there is no enemy that is gauranteed to have access to this effect.
Armor- you may not need any if you mix Stealth with Illusion, you could choose Alteration or Security instead, both for getting into locked doors, Alteration has some combat effctiveness, shields mostly, just ignore the burden effect, enemies never carry enough for it to do anything.
Marksman- or Destruction.
Mercantile- I prefer Illusion, if you decide to use both Conjuration and armor put Illusion here.
Sneak- central to the concept of an Assassin.
