-You should play all of the 3 houses (not possible in one playthrough)
As for being completionist, you should still give the game a shot. I am more of the collector, than completionsit type, and pretty much all of my characters had incredible collections of all unique items etc. availabe to me at said gameplay.

but if im not mistaken (havent really played these lines in a bit) fighters guild asks you to kill the head of the thieves guild and the thieves guild asks you to kill the head of the fighters guild? perhaps theres still a way though...
TIP: You should get some infos about the master of the figthers guild and the man in the guild in ald'ruhn
The two most priminent vampire quest-locked missable items are the Darksun Shield and Spellbreaker. You need to be in the Aundae clan to get The Vampire Hunter quest that gives you the Darksun Shield. Do not use it until you're cured, it WILL kill you if you're a vampire when you equip it.
Spellbreaker is obtained by finding a vampire trapped in the Salvel Ancestral Tomb that, only if you are a vampire yourself, sends you to Bthuand to obtain it. It will not be there if you're not on the quest.
If you are not using the unofficial patch, then Eltonbrand is exclusive to vampirism as well. Getting it is obscure and requires you to already have Goldbrand, and then to have exactly 11171 gold on you as you turn in the vampire-only quest Shashev's Key for Sirilonwe in Vivec's Mages Guild. If you are using the unofficial patch, then having exactly 11171 at any time, vampire or not, after having Goldbrand will give you Eltonbrand.
While you can get it any time, vampire or not, there is a vampire quest you will probably be pointed to through dialogue where you will obtain Marara's Ring, one of the best rings in the game (Constant Effect - Reflect 20% on Self, Fortify Acrobatics 10 pts on Self, Resist Normal Weapons 40% on Self). It is not missable but you're very unlikely to find it, unless you know exactly where it is, without the vampire quest related to it.
Vampire as mentioned above. Similar to the Great Houses, you can only do one bloodline.
And the Fighters/Thieves Guild have conflicts when it comes to Sottildes Code book that can prevent you from joining each other. Both quest lines would have you destroy the other near the end.
My guide walks you through getting the latter without issues. There's multiple ways to resolve it. If you need the Join All Houses mod, I can provide it to you. I don't know if it's disappeared from the internet.
Finally, for Solstheim (Bloodmoon) -- you have to choose between Werewolf or Skaal. Both are excellent storylines (but you can save before the choice and do both). If you do choose human, you can get a ring to allow you to be a werewolf at any time after completing the quest.
And you can join 1 great house of 3.
You can just play the game and roleplay your char, best to play this way and then locked content because of your choices are good for roleplay
Well I still have my whole life ahead of me so I'd think I could do it. Some people might play only one game, a mmorpg for example, for years so what's the difference?
