Large Combo - Tons of points, before Pizza Time, try and rack up the kills even if you have to take a detour. But watch the meter, Can't P-Rank without keeping the combo the entire level.
Toppins - Optional, but very valuable to go for.
Lap 2 - Mandatory 3000 points for free, usually high-enough to boost to an S-rating.
Collectibles (including pizza time clocks) - Low scores all around, both big collectibles and little ones. Best use is for saving you from dropping your combo.
Secrets - Only here because they're mandatory, has quite a few collectibles, so the score is still worth it.
Treasure - Always gets you a good chunk of points - usually enough to instantly A-Rank it.
Also, try not to hurt Peppino. One or two times is OK, but if you see the messages about his or Gustavo's sufferings, then you might have a bad time.
I guess that makes sense. Probably to prevent the player from starting a combo as late as possible and encourage them to start it from the get-go.
