Upstairs 2 empty room and one is closed.

I assume you got the necessary info from the innkeep. Have you tried waiting for another hour? If I remember correctly, I also had some small issues with making his nephew appear. It's the room on the left side (in the center) when you enter the tavern.

Yes,i tried several hours.Nothing change.
Barkeeper has always same response:every room has already customer.

Sorry, but in that case I'm out of advice unfortunately. Did Helen/Mary ask you for quality time when you entered the tavern and before you could talk to the innkeep about Levasseur's nephew? Sometimes that can break quest related stuff to happen as intended.

There's another room on the 2nd floor. It's near the table that's right above the bartending counter.

Several reloads earlier saves no help.

I don't understand what you're saying.

So there are 3 doors put side-by-side. Those aren't the rooms you're looking for.

The correct room is the guest room that the player will stay in whenever he asks to rest in the tavern.
If he still doesn't spawn then send your savefile as a bug report.
