I started this game as a demo, played four-ish days before buying, does that matter??
I'm not sure I understand exactly what might be gate-keeping you here; there are two possibilities.
There's a 'weird . . . glowy stone' on the western side of the map northwest of where the fisherman dude is; that's not the first mine but rather something that unlocks later. The first mine you need to enter is found at the very eastern end of the beach in the very southeast of the map.
If instead you mean that you've entered the mine (in the very southeastern side of the map) but can't get to the second floor, then all you need to do is break all the rocks in that first room; one of them will open the door leading to the second floor. Once you have enough resources, you can craft a seal-thingy and permanently unlock floors by installing seals in the font-looking things; crafting the seals isn't necessary to progress in the mines, but rather to progress that specific quest (although they do make reaching the later floors easier).
(Apologies for all the technical jargon, sorry.)
If I've misunderstood, let me know and maybe post a screenshot of your quest log/the location of the 'weird . . . glowy stone'.
