scrap the Artifact "Killing Gaze".
Those Enemies are immune for a short time after spawn and won't cut it.
You definetly want "Hastening Sands", the rest is up to you.
Restless Wheel is nice too for faster Agony Ramp Up.
You can pick any char you like, but you definetly want to use Ring Blades or/and Arcane Splinters. Those Abilites have the best screen coverage and kill everything on spawn,
which gives you lots of kills.

Sage is amazing with Ring Blades as main Weapon,
Swordsman and Warlock are easy too, since both have strong main Weapons with lots of coverage.

This quest was even more ridiculous before, you were supposed to kill 66,600 enemies, not 50,666. Closest I got to that was 62k monsters but I also struggled with it.
Are the spawn numbers that great that losing 10 minutes is outweighed by them? If so, I've been doing this wrong.
The key is to use the sage and only get 2 abilities (ring blades first, phantom needles second). If you get more more abilities than that, the constant spam of your abilities all over the screen causes the game to spawn less enemies than it otherwise would.
I followed the "super smash homies" build from this guide https://steamcommunity.com/sharedfiles/filedetails/?id=3172296902
and used the phantom needles traits build from the same guide's "critical mass" build.
I finished with ~5 seconds to spare.

Sage with Vision Crown boosting the range by +100%, Wooden Ring +100% Crit Chance, Iron Ring with Growth for some additional damage, Hunter's Garb (Boar Power) for even more damage with Pace Setter (Unrest) and Quickhand Gloves (Boost) increasing attack speed made this easy.
Ring Blades as the "main weapon" filling up the whole screen was silly.

I don't have all the items yet, only 50 hours in, so my build is based on what I have. Crit Bonus would probably be better considering all the attacks are crits to begin with. Archer mark to further boost the crit rate and bonuses through Weapon Proficiency.
There's probably plenty of ways to further optimize the damage the build does. I've been using it for the Berserker III achievements that I've been doing now to complete map specific ones.
Followed this and managed to get it first try too. Once you get enough upgrades and agony up it just goes up very fast.
