One thing has in fact changed about my setup. Last time I played I used my audio interface for my guitar, but audio output was my soundcard. These days I run both input and output through the interface. For whatever reason, RS doesn't like that and won't detect the guitar.
The problem can be "fixed" by opening Rocksmith.ini and changing ForceDirectXSink=1 to ForceDirectXSink=0. Don't ask me why this works... I don't even know what the setting does. Found the fix through trial and error. :p
