So don't expect plug and play. For Scarlett there are some guides made.
There's also a guide for Behringer, which I believe is a cheaper version. But getting the right legacy driver files can be a problem sometimes I have heard.
A interface in general works best with ASIO, k not all support that....)
you would need to have mods for that also, like RSasio and to complement it with RSmods. It does bring it's own problems with it though if you also want to stream and/or use discord since ASIO only works well with audio exclusivity, although there are workarounds but it does need more fiddling with software.
Also in RSmods there's a hidden function unlocked for not having the need of the RT cable by choosing direct connect option. Might try that first.

ASIO is designed to be in and out on the same device so you get the lowest latency possible.
If an audio interface doesn't really support ASIO, or one can't find the legacy drivers some tend to use ASIO4ALL, but better not do that. It doesn't use ASIO but impersonates it and actually uses the windows audio protocol, so it doesn't give the same benefit. FlexAsio could be better then.
Also it can not be used next to the RT cable unless you either remove the RSasio files or add another program like voicemeeter that can split sound sources, it's not always easy to configure so it does add to the bunch of difficulty in setting things up. And an update can mess it up too.
The no cable patch is a very old hack, while RSmods adds also more other benefits.
Do you perhaps also use the "beta" version?
It's these days pretty redundant.
If so, the mentioned mods must probably don't work since they are updated to today's versions, or you should try out different older versions of those. But troubleshooting can be very difficult if something goes wrong.

Run the installer, it installs itself in the Rocksmith folder, there's a one button action to get most things sorted.
it might ask for a path to save backup profiles, for that it opens file explorer and you guide it the required path.
For best results Rocksmith shouldn't be on.
if you open RSmod you will see a lot of options, choose what you want to have, like a quicker startup if you have installed on a nvme drive. exit button, Oculus fix, color changes and even a tone switcher so you don't always have to Retune or not that much for different songs.
