Make sure the RealTone cable and your sound output device are both set to 16-bit @ 48KHz.
Try working through the audio setup guide that comes as a PDF with the game (or other audio configuration guides on this forum and the official forums).
The problem was solved by accessing a notepad file(one of the game files) right click on rocksmith2014 in your steam ,my games. Browse Local Files - find all the files that were installed with the game- on mine (PC version) the file I needed to modify was called rocksmithconfiguration - open that - it should be a notepad file with a list of things and = signs, like; win32ampreference=2 or something etc.etc.
It was just a matter of changing the numbers after some of the = signs and everything has worked fine from there - I got past the tuner and more importantly, when I strum a string it is CLEAR and actually sounds like a distinct note, not just static - so thanks to the guy that helped me solve my issue - I.R. Dianetics - he is a pure genius !
Well, thanks for being so specific about that.
