Using Audio interface over real tone cable
I got RS_asio Setup and i get sound but i cant get pass the calibration meter , getting sound but says i need to hit harder but im stuck on this screen, Any suggestions? i got volume maxed out on my Bass and tone up all the way.

On my MAC there is a global setting in the sound preferences for input volume and this can interfere with Rocksmith's detection. Could this apply somewhere on your system?

Also early on in my Rocksmith 'career' I realised my pickups were way too low and this negatively affected the guitars output. Too high however and the pickup magnets pull the strings and make the note pitch waver which also messes with Rocksmith's note detection.

an internet search tells us how to set an optimal height. A good starting point in the UK is the depth of a 5p coin; my pups are lower than that however - in the US it's a dime or cent or somesuch - google it! :-)

Check and raise the cable's gain. Windows likes to reset it to a default of 17. This will usually change to something higher after you calibrate. Crank the sucker up to 75 or so and see how that feels/sounds.
