You could test it by playing Blitzkrieg Bop a few times and see if it drops into MasterMode properly...
Some songs seem to really hold back on going into master mode.
One missed note and it could hold off until the entire next verse is cleared without a mistake.
You just gotta hit every note just right on some songs.

Found two answers that are somewhat contradicted
Go to riff repeater, select the whole song. Run it up to 100% difficulty. Ensure the Master Mode = yes. Then you'll have to play through it several times before it starts to enable it.
Or
Start playing the song in Learn A Song mode

Go into Riff Repeater and set the difficulty to anything below your mastery level (I put it at 90%)
Hit Resume Song and play it through to the end
Play the song again and Master Mode might kick in with a bit of luck

If you really want MM, then go into Score Attack and choose Master. No questions then (except you might have to unlock Master by first fully playing Hard).
