It could also be worth ending some other programs in the background specifically audio programs. Rocksmith wants full control of your sound systems to perform its best. If you want Rocksmith to stop stealing control of your mic, right click your sound icon in the taskbar and select recording devices. Choose your mic, then go to the advanced tab. Uncheck "allow programs to take exclusive control of this device" it can bugger things up, but it should allow you to talk to your friends while you play. In the game there is also a setting for audio exclusivity. You might have to turn that off too.
Alt Tabbing can be a bit hit and miss for most games. I don't have much to offer other than the same advice above.
