Go into your system preferences (you do not need Rocksmith or Steam opened/running for this). Choose the "SOUND" preference pane. (looks like a speaker).

On the "Input" tab, you should have the Rocksmith USB Guitar Adapter selected... and you should be able to see it's selected Input Volume and Input Level. If nothing is showing up, then you have a bad cable. If it's showing up (like mine) maybe you need to turn the input volume up.

Win 10 privacy issue

settings/privacy/microphone allow apps to access microphone = on

Initial setup FAQ

N.B. Sometimes sound settings can pose a problem when set to Surround or like that. You might also try to disconnect other equipment to see if that could create a problem in the setup.
Also if possible you could list your Rocksmithi.ini here. It might (on a windows pc) be in C:\ProgramFiles\Steam\SteamApps\Common\Rocksmith2014/ or like wise dir with (x86)

The last link goes more in depth about it too.

Same problem on my mac: After installing the Rochsmith Catalina update (but not updating the OS), the realtone cable doesn't work anymore (neither does mic as input). But in the system settings I can see that both audio sources are working properly. But Rocksmith is completely muted :-/

Rocksmith Driver Issue here since last windows update.
Privacy Settings are "microphone use on" for Rocksmith (that solved the same problem with earlier windows updates).
Driver not working, no driver update possible, windows says "not possible to start (code 10)"
(Eigenschaften: Das Gerät kann nicht gestartet werden. (Code 10), Treiberfehler)
