How to tune a guitar with locked tuners?
I just bought a Ibanez GRG 270, and apparently it's not tuned well and Rocksmith won't recognize it, I tryed tuning a bit and apparently I heard something happening with tuners so I'm assuming that's not a good idea lol. Do I have to take them off just to tune it?

I believe you're referring to the double locking tremelo, aka Floyd Rose. I also use a guitar with a Floyd Rose at times with RS, so that shouldn't be an issue. If it's not picking up the strings when they are played then make sure that you've gone to the calibration and have it being picked up. Additionally, take a look at your pickups and see if they are too far from the strings. If so, you can raise the pickups bringing them closer to the strings so that they are being picked up when you pluck them.

Sounds good. If you start having issues with the strings not being registered correctly when you know that you are playing the correct notes, then it may be an intonation issue. You can look up how to raise the pickups as well as how to set the intonation on youtube or the web. Just look for your guitar's model when you do so. You may not find your exact model, but you should find a similar guitar. If you don't feel that you can take care of the issue yourself, you can either take it to a shop and have it done for you, or ask a more experienced guitar player.

Google how to tune an FR type guitar.

Also how to setup one up.

It is not hard to learn, and there will many methods to choose from. Find the one that most fits your guitar and one that works for you.

If your tuning machines are locked you should unlock them.

Did you have the trem set level with the body? I'm wondering if you got the strings too tight. They're fiddly to tune but once you do it they stay in tune pretty well. If you're going to use the game mostly for Rocksmith, you can either block the trem from the back or get a gadget called a Tremel-No. Either of those things will basically lock it like a hardtail, the Tremel-No allows you to unlock it with a thumbscrew.

I wouldn't say dumb. It's just part of learning when you're new to FR. As TOK mentioned, unless you have your reasons for wanting to use the FR, I'd block the back of the trem. There's lots of stuff on the web on how to do this and a variety of videos. Here's just one:

I understand. I have several guitars, (I know, that's unusual for a guitar player :steammocking:) and I sometimes use my FR with RS and IDK, it's Ok IRL, just kidding. It's been fine for me. I'll switch guitars though if I want to play really low tuned stuff. Anyway, Have fun.

Its totally do-able to use a Floyd wtih the game. My main Rocksmith guitar is an Ibanez RG450DXB with a Floyd. I rarely have to do anything with it for Rocksmith now, but when I do its usually only a small tweek of the thumbscrews at the base of the bridge.
