For the mic issue, that's hard to say. It could be a variety of issues and I haven't had that happen to me. To help, I tried to find others with the same/similar issues. Here's some links with their suggestions. I hope this helps you.

It was the first link with a few different suggestions to try. I'll post them here.
1. This could be an audio problem than a tuning problem; if the audio input isn't working properly then there's no way the game can recognize notes.
Make sure the RealTone cable and your sound output device are both set to 16-bit @ 48KHz.
2. Some people have had problems if they didn't calibrate their guitar right. Choose to recalibrate again. As I understand it, it is best to play as loud as you can. Then when it tells you to silence the strings, only mute for a moment. That way the noise gate gets set to the sound your strings make when they aren't being played.
3. If the two above don't work, try accessing a notepad file(one of the game files) right click on rocksmith2014 in your steam ,my games. Browse Local Files - find all the files that were installed with the game- on mine (PC version) the file I needed to modify was called rocksmithconfiguration - open that - it should be a notepad file with a list of things and = signs, like; win32ampreference=2
