How and when to get into the expansion contents?

Can someone explain that to me? Does the expansion expand the main storyline? Are they related?

If you talking about Iki island You could do it right after it available, but you could see some spoilers there (not sure if they removed lines)
If you're talking about legends. It's not related to the main story and it won't spoil you the story. You could play it whenever you want. It's separate MP game with separate progress.

I don't play MP, so how do I get into Iki island? Do I have to pass a certain main story mission to unlock it?

on ps5 it was usually right after act 2 started. there would be a mission 'journey to the past' in the west. I've reached act 2 and it doesn't show up for me just yet. guess I'll be playing some more main story until it shows. but no there are no real spoilers. you can just hop into iki island no worries

after you finish act 1 and unlock the rest of the map a marker will show up :)

Canonically its between acts 2 and 3. Really you just want to beat it before you beat the main story so it makes sense
