No, gonna go through my first ever play-through normally, with English VO. Will most likely do a second run in Kurosawa and with Japanese VO.
The Samurai Cinema feel makes replaying the game feel not repetitive. In addition, some combat elements and a lot of interaction with the world are a bit annoying without the color cues in normal mode (particularly any time you have to find something). That frustration is lessened if you've already done it once.
Ah, then still no. I didnt go B&W in LA Noire or the washed out colors in AC Mirage, keeping the regular colors. But understand why devs put in things like these.
