There are three co-op modes in the base game (four, if you count Agent Hunt, but that one pairs you up with random people and there is no way to control whose game you end up joining, or who you pair up with as a co-op partner): Campaign, The Mercenaries, and The Mercenaries No Mercy.
Campaign co-op is pretty much self-explanatory. There is no actual weapon customization around in RE6 (sadly). Instead, you have a skill points system that you can use to purchase skills. You can have three skills equipped for a skill profile at one time, and can rotate between 8 skill profiles even while in-game through the options menu. The skills can range from anything like allowing you to reload quicker, to removing your laser sight/reticule in exchange for having your weapons deal more damage. There are quite a variety of these different skills, and a quick Google or look at a Gamefaqs walkthrough will probably turn up a full listing.
Campaign has five difficulty levels: Amateur, Normal, Veteran, Professional, and No Hope.
Amateur is, as expected, the easiest and least challenging of the difficulties. Enemies are overall weak and the game automatically performs most cinematic QTEs for you (though this feature has become redundant with the patch of an Auto Action option, that can be used for all the difficulty modes).
Normal is a slight step up, but still pretty easy.
Veteran is where situations start to require a bit more thought. And an intermediate knowledge of the game mechanics is suggested.
Professional is yet another step up, and is quite punishing in terms of how much damage enemies can deal/take. This is where using cover to catch your breath and plan your next move comes in handy. Starting with this difficulty, enemies do no not drop health items, and the only ones you'll find are hand-placed. Ammo is slightly more scarce.
No Hope is a difficulty that was added in a patch later on. This mode demands patience and an intricate knowledge/mastery of the game's mechanics. Enemies can put you into Dying with only two or three hits, and getting hit by even a few bullets from Ja'vo is likely to send you into a stagger animation and make you lose a bar of health. Worse yet, ammo is generally much more scarce and most of the time you'll break open boxes, only to find skill points (which are doubled in worth for No Hope). You are also not allowed to equip skill profiles in this mode. Worse still, enemies do not occasionally drop herbs or F-Aid Sprays. The only place you'll be seeing these items are in hand-placed parts of chapters (which are few and far between). Avoiding damage is key, as the game will not bring you back to full health upon going through a checkpoint and dying like on the other difficulties. It's not uncommon to find yourself barely getting by on one bar of health for the whole chapter. There is quite a lot of resource management in this mode, and you will likely end up finding yourself running out of ammo towards the end of each character's respective campaigns. Personally, I find this mode to be the most fun/tense of the difficulty modes/game modes (especially for Chris and Jake's campaigns), as the game punishes even the smallest of mistakes on the player's part with the chance of death.
The Mercenaries sees you killing enemies up to a combo of 150. The goal here is to keep the combo uninterrupted until you reach that number and before the time runs out. You start out with a time limit of two minutes, but can find time bonuses on each map to increase the time limit. There are also combo bonuses that you can find on each map to further increase your score. Time can also be rewarded in increments of 5 seconds (for a standard melee kill) 7 seconds (for a coup de grace) and 10 seconds (for a counter that delivers a killing blow), and this will probably be your best bets for increasing the time limit. You are also allowed to use skill profiles (though the amount of skills you can use for Mercenaries is a bit more limited in selection than what you have for campaign, and you can only equip one skill in each profile. You are also unable to switch profiles mid-game).
The Mercenaries No Mercy is a mode exclusive to the PC version of RE6 which greatly increases the amount of enemies that are spawned. The combo is also increased to 300, rather than 150. I find this mode to be a bit too mindless to play consistently again and again in one sitting, as it's way too easy to keep the combo going and you can spray and pray into enemies (which are weaker to compensate for their increased numbers) with little regard for ammo. Zombies are more challenging on this mode, while Ja'vo are easier. You are also able to play with Left 4 Dead characters in this mode, and there is an option to have Tanks and Witches spawn at certain points in the match.
