Just think of things as a gigantic bureaucratic, galaxy spanning nightmare to get yourself into the lore. Aliens are bad, mutants are bad, Heretics get burnt at the stake and humanity is where its at, the Emperor is our loving god/father figure that wants to save humanity as a whole.
You dont have to read 50 books and have played the table top to get into it, its a fairly self contained story and you get lore clues as to whats going on, theirs highlighted sections of dialog that give you snippets of lore that should carry you through confusing parts, not to mention the forums where basically everyone here would love to explain any confusing lore detail to you.
40k lore is both deep as an ocean and shallow as a puddle depending on how much the universe "calls" to you and sucks you in.
Humanity owns the milky way, its beset on all sides from enemy's within (Heretics) and without (Xenos/Aliens). Aliens are untrustworthy and use/abuse humans at every opportunity. Mutants are probably (99%) Chaos spawned abominations that also want to destroy or use regular humans to achieve their "Gods" goals. Heretics are madmen that want to see the galaxy burn in the name of their dark "Gods" (The 4 gods of chaos)
Seriously, just remember: "Kill the Alien, Purge the Mutant, Burn the Heretic" and you should enjoy it. The game is pretty expansive and lets you explore lots of options if you are down for multiple runs exploring different sides of the overall wider "lore" story (Xenos lover, Chaos, Puritan, Regular Joe etc etc etc).

Easy enough to follow then by what you've typed. I suppose it'd be the same for me as it was with the Pathfinder games, which again, I had no experience with but managed totally fine and furtherly enjoyed.

It is VERY importent that Warhammer 40k takes everything to the extreme. If you take that stuff serious you will get into a weird mindset. Warhammer frequently crosses over into the satirical realm while playing it off as dead serious. Don't take it dead serious.
