When am I locked in to completing the main quest an unable to do side quest here

The chapter ends when you go to a spaceship. Either Yrliet or the inquisitorial agent on Dargonus will send you there (depending on if you recruited her).

Chapter 2 ends with your inthronisation on Dargonus. I recommend to finish all quests before that, especially on Footfall, which gets partially cordoned off afterwards.
After the cut scene with badly drawn black-and-white figures you are in chapter 3. You can no longer access the traders then until chapter 4.

When you have to go to Dargonas for a big party you should make sure you've done everything you want to first.

That sequence is actually in Act 3. Act 3 starts after the party finishes.
