Your Space Marine can identify as a man, a woman, an orc, or even a Warkopta!
They deal with chaos uprisings, not xeno invasions.
That being said, they are already featured and playable in Gladius: Relics of War, Inquisitor: Martyr and Rogue Trader RPG.
They wear power armors, but they are orginary humans, just little too much into believing Emperor to be actual god.
The main goal is to protect the IP from destruction, like many others before them.
For 40 years, Space Marines were male because the transformation process required a young boy's body. In Codex Edition 3, it is mentioned that some modifications enhance the Y chromosome and boost testosterone production, but only 2/3 of men can withstand it.
The selection process is extremely challenging for females, although it varies by Chapter. For instance, the Space Wolves recruit heavily wounded fighters from war battlefields who have shown exceptional fighting spirit.
Also it reduces other factions where Female or others are representat.
Eldarf have a Lot of Legendary Female Fighters.
Even when i Male Took the Armor, the Female Soule would take over and transform him.
Orks are ASexual, for them Fighting is Sex
Tau, one of the biggest Commanders is a Female
There are Factions where are only Woman.
ASK for a Sister of Battle Faction and i would even agree.
These groups cannot be swapped out for player taste because units would be radically different Marines and Custodes different grades of super soldiers in power armor, sisters are religious nuts in power armor, Guard are normal humans in a t-shirt.
When Tera or a Sanction Planet is under Attack, the Sisters would not hold Back.
Flames heal against Tyrannids.
There is a conversation between The Emperor (The human leader/god and single most powerful psyker(mage) to exist) and malcador (gods BFF and immortal) wherein Malcador tells the Emperor that he believes the Primarchs (the Emperors non biologically related super children) should be female. the emperor laughs this off and either does or pretends to believe it was a joke... it was not.
So the Primarchs were made male, and thus the Space Marines whom are made using the genetic material of the Primarchs must also be male.
unfortunately the ability to make more Primarchs female or not has been lost as the Emperors body was wounded seemingly beyond repair and has been stuck on a giant throne that acts as both life support but also a physic beacon for warp travel (light speed but through hell)
The existence of Female custodians (Even more super than space marines) does not lend any in-lore credence to Female Space marines as Custodians are NOT made using geenseed like space marines are, they're simply normal humans pushed to the absolute natural limits the human body can be pushed to without genetic alteration.
In my honest opinion, we're going to eventually see female space marines we've recently had female custodians be confirmed and while as I said that means nothing in lore on the subject of female space marines it does show that GW is moving into a more inclusive mindset... and that's something 40k fans are going to have to get comfortable with.
I for one don't really care, so long as female space marines don't behave differently to a male space marines, both mentally and physically.

That can of worms aside I'm sure players would love to play as Sisters of Battle, or Sisters of Silence, who by definition are all female, and if they were implemented with full customisation options I'm sure it would go down well. I'd love a game like that myself. A full campaign with the same mechanics but as a Battle Sister fighting a Chaos or Genestealer cult somewhere - yes please! Imperial forces often find themselves fighting in the same arena so that wouldn't be an issue to have them fight side-by-side.
However, the game is called Space Marine so I doubt the devs have this in mind. There's lots of room in the lore for female characters and great games showcasing them - the two factions mentioned above, the Inquisition, the Imperial Guard, planetary rulers/governors, assassins (most Callidus type assassins are female because of similar biologically based lore reasons for example), hive gangs (the Escher gang of Necromunda are all female for instance) and xeno races like Tau or Eldar. In the lore people can get modified with all sorts of biological or mechanical enhancements which may not make them full on Astartes but certainly enhanced super-soldier types etc though that may pose a risk of being labelled as heretical in some quarters of the Imperium.
I've not played it yet but I'm really looking forward to the campaign - it's a shame all things today become embroiled in identity politics and people can't just love a thing for being itself but that's them internets. I hope you enjoy it!
So my question about including other factions is a little redundant but possible, although the game is centered around specifically the soldiers who were genetically altered using the Primarch. So playing as other factions would be better suited for another game then this one.
While I would prefer to play as a female, that lore makes sense and as previously stated, I'm still willing to play.

If you're interested in the lore/universe more there are factions/groups that feature females. some of which have been mentioned by others.
