If it doesn't work,try to set the application to "Windows 7 Compatibility Mode",that can also fix the problem.
First time it happened I tried doing a verify game files, which did make the game work once. But then the next time it did the same thing. I really don't want to validate the game each time as it takes around 15 minutes and it's not a guarantee that it'll work.
Other fixes other people suggested like putting the xlive.dll in the directory didn't work... because the game doesn't use Games for Windows LIVE anymore unless you change it to the beta branch and I don't want to mod the game (as I said it works fine on my PC and my laptop and they're both stock with no fan-patches or mods or anything).
Anyway, the usual fix for something like this (a game not launching correctly) is running it as admin. So open the local files, right-click re5dx9.exe, Properties, Compatibility tab, and check Run this program as Administrator. Now the game works fine, the only weird thing is that CMD window opens up with "res = 1, err = 0x00000005, Access is denied." and stays open indefinitely. This doesn't seem to affect the game in any way. The only other thing is that unless you close this window, Steam thinks you're still in game so if you care about your in-game hours make sure you close it.
Another suggestion I would make is if you get the game up and running, cap it to 60fps in the video settings. Having it uncapped lead to some weird stuff like unkillable bosses in online co-op.
The CMD window points to Launcher.exe in the game's directory. I assume this is some sort of wrapper or workaround/replacement for GWFL. I don't have this set as Run as Administrator but I'd try that too if re5dx9.exe doesn't work. Hopefully this fixes it for you.

Change Compatibility to Windows 7 for both Launcher.exe and re5dx9.exe (didn't do anything, so I changed it back to default).
Another thing I did was delete the game's config file. Default location:
C:\Users\[username]\Documents\CAPCOM\RESIDENT EVIL 5\config.ini.
Maybe this had something to do with it. You'll have to reconfigure your video settings after deleting this.
Another thing I did was launch re5dx9.exe directly from the directory rather than using Launcher.exe like Steam does.
I also reinstalled the game but that didn't fix it either.
