Memorising the track layout is always helpful as it's the same every time you play, just like any other racing game out there. You may need to replay the level multiple times to know the layout and get a feel for it.
Every 20 rings gives you a speed boost which you can activate with the action button, though I would only really use it for straightaways where boost works best, though you can take risks if you're that kind of driver.
Drifting is also useful. By letting go of the accelerator and quickly holding the accelerator again, you'll be able to drift by turning, allowing you to make sharp turns more quickly, which can also be useful for missions 3 and 4. Remember that these play more like an arcade racer.
Hopefully these tips helped you out at least a little bit. The A rank requirement for Route 101 is under 2:45.00 for mission 1, and for Route 280 it's 3:20.00 for mission 1.
