Apparently the mutations that get "cleansed" are the most recent ones added so it's easy enough to manage the bad ones. I can't confirm this 100% as I'm lacking the necessary things to test it myself but from what little mutation stuff I've worked with it seems to be the case. I would still save beforehand just in case.
From my (admittedly limited) experience with normal mutations I've had it take the most recent ones as well. That one is easy enough to test with enough mutation/cure mutation potions/spells.
The formula can be found on the wiki's page on Riding, and weight doesn't factor in.
It's just that the horse is pretty fast, and thus requires either the horse getting more Strength or you having a higher Riding skill to offset the penalties of riding a fast mount.
Unless it's been changed giving ether equipment to companions 100% gives them the disease.
Ether wing (can be upgrade to double wings if same mutation happens) privides tons of speed
not sure whether it works on mount or not.
