You don't unlock Prestige Classes. You can only select them during a run, if you meet the requirements. For example, you can always become the "Champion"-prestige class, if you've spend at least 25 points on martial-powers.

You can view (and select) prestige classes on the same screen where you select or level up your powers. Click on the square with the + at the top of the screen.
You don't unlock prestige classes. They are all available, however you can only take them if your build meets certain requirements. For example a common one is 25 glory spent on skils in x or x and y.

Some have other conditions, such as the transformation based prestige classes require you to either have a specific helmet or a piece of gear with over a certain value.

If you select powers at the screen where you select race, class and religion you can see all the prestige classes and what conditions need to be met to pick that.

I was quite enjoying the horrorform prestige class... until a gold demon swallowed me. That's life, I suppose...
