How big is the DLC?
Is the Mea Culpa DLC in any way comparable to the combined playtime of Blasphemous 1's free content updates?

No, Blasphemous 1's free content updates were much more substantial, ironically enough, as they were free.

2's DLC includes 2 new areas, 2 new bosses and 1 new weapon which is a direct upgrade to Ruego Al Alba (the curved sword). 2 existing areas also got expanded, and there are some new altarpieces to collect. The DLC is overall VERY platforming focused.

Honestly, wasn't a big fan of the last boss. It's like Eviterno if he was actually unfair. Extremely aggressive with very large hitboxes, forcing you into constant parries with barely any windows to land hits.

Yeah, I've read some of the reviews. Is there an overhaul planned? I wanna see the end of the story, but the content to price ratio (especially after all of the free additions of Blasphemous 1) feels hard to justify. Maybe I'll wait for a substantial sale.
As a die hard Blas fan, not really I hate to say. The return of mea culpa was great and the new platforming is fun and can be kinda challenging, but its nowhere near the size of say, eventide from 1. I don't get the complaints about the last boss tbh it's probably the most fair and rewarding fight in 2, but wait for sale if content is your thing. Will say it has some of the best music they've made for the series to date, especially in one of the new areas Santa Vigila

The map is greatly expanded and there are lots of platforming challenges if you enjoy them. I had lots of fun. It is not so focused on bosses compared to the expansions of B1 but has more of the rest.

Has anything been said about a 2nd DLC?
