What are the best Towers?
In my 4 slots I usually have a "Focus Beam", "Friendly Balls", "Missile Turret", and "Slow Mines".

What is the optimal tower combination?

All depends on your play style and the weapons you're using.

I find Focus, ACP, Range & Slow Mine to be a good all round combination, I know a lot prefer the Violator to Focus but I guess that's preference. If playing multi, a mind control tower near the ACP works wonders but I wouldn't sacrifice the Focus tower to bring it in solo play.

I like to run a variety of damage towers myself. Lightning, Focus, Scatter Laser, and Cannon, because I run co-op with a friend who's in love with the various dispensers and buff towers. It's all fun and games until I'm permanently stuck in slowfields as Haigan, heh... Though, I'm not sure if those are more annoying, or constantly getting Rex Missiles up my butt is, since he loves Sweet... The amount of times I've been sent flying... *sigh*

I go with Focus, Gatling/ACP, Range Spire, and Slow Field.

I take Focus, ACP range spire + something else. I'm not sure about my 4th tower yet.

1) Depends on the map (particularly whether there are a lot of flying enemies and/or bosses), which other towers you're taking to complement them, and your character and perks.

2) That being said, in general the gatling, violator, and rocket towers are the worst. The scatter laser pretty much always trumps the gatling, and the orbital strike relay pretty much always trumps the violator and rocket towers, which you can see both from the numbers analysis in the wikis, or just from playtesting them yourselves.

3) Cannon vs focus really depends on whether you've activated the +HP and +regen feats. If you have neither feat on, cannon is better. If you have both feats on, focus is better. If you have just one of those feats on, it's a toss-up. Makeshift cannon is also an interesting alternative to the cannon and focus, regardless of which feats you have on, but it's a bit of a game - stunning a mob is great, but accidentally deactivating is not.

4) Lightning, ACP, and slow field have the potential to be great, but only if they're being boosted by a range spire. If you or a teammate aren't bringing a range spire, lightning, ACP, and slow field simply won't affect enough mobs to be worth it.

5) The anti-air tower is the first and final word against flying mobs. It's so cheap and effective, and its damage scales up so quickly with overcharging, that if you just put one of them in the middle of a map, even without a nearby range or amp spire, it will handle almost all of the flying mobs for you, freeing up the rest of your towers to concentrate on the ground mobs.

Gatling has the highest DPS and the highest DPS per resource in the game.

Focus at least for survival because overpowered with damage amp it hits so hard only tower that competes is acp within a overpowered range amp. Because acp can hit every single enemy at once. But do not underestimate mind control will quadrouple the time people spend within kill zone of towers if placed correctly. Slow fields will double the mind controls effectiveness by preventing as many smaller enemys from running into mind control towers range and stealing the mind control buff.

Gatling is most effective dps per resource till armor shows up then its completely useless. So in survival when mobs get 10+ armor (-3000 per shot)gatlings shoot fast but low damage meaning each shot is mitigated. Also as enemys have more and more hp it means focus will have to target enemys longer ensuring each focus beam reaches maximum potential. As each tick of focus gets stronger up to 5x original damage.

That's only if you don't consider range and armor. Gatling does little to no damage against armored enemies, and its range is smaller than the scatter laser (especially at level 3), which means the gatling's DPS is actually 0 for significant periods of time, while the scatter is still hitting mobs. Numbers on paper aren't everything; playtest a stage twice, doing everything the same except using gatlings one run, and replacing them all with scatter lasers the second run. Scatter lasers invariably trump gatlings in reality.

Depends on many things. Most of all if you're playing solo or co=op. I find solo impossible becaue it's a different game to co-op and needs different mazes and towers.

If we are talking damage wise it will be the anti air. Most of the towers have a good niche though.
