It's easy to die on a stupid mistake, or on bosses for which you have forgotten the tactics. I'd recommend running a New Game + and getting familiar with what's coming ahead, backing up your saves and/or leaving before it autosaves to train, and then run the I am the Night mode once you feel comfortable with it. Doing it in one go is near impossible imho, just like however experienced you are, you can easily break a combo in challenges because you pointed not exactly at the right ennemy, waited a tenth of second too long before your next move, get caught in a combokill while an ennemy attakcs you, and so forth.
WB games really improved on their combat system with their LoTR games which is much more forgiving combo-wise, but still maintain a good challenge.
As you probably know the game autosaves and doesn't let you replay certain parts, but there is a way around that by copying the saved file to your desktop and then putting it back into the same folder when you want to replay a certain part - Arkham Origins saved games are located in your Steam folder - mine is
(C:)\Steam\userdata\(unique number here)\209000\remote\(your saves)
Practice on New Game + (cos that's what I Am The Night is only with 1 life) and when you get up to parts you want to practice, like when the Bane rooftop fight starts or when the firefly fight starts, exit the game and go to the folder above and copy/paste the save file into a folder on your desktop, and whenever you want to replay those tough parts in the game you just copy/paste that file back into the folder above.
* and remember to swap it with the one that is already in there, rather than overwrite it.
