1. no, they stay in that area. I do know that sometimes the lighthouse and the plaza are accessible for them in one place ( for example if you put the guest house on the lighthouse and your entertainment area are in the plaza, the guest can come down from the lighthouse to your plaza )
2. they matter in a way, because sometimes your farm will get attacked by pests ( i think their level goes higher if you follow the sujimon substories) and if you dont train your sujimon enough then they cant defend your farm from them

In regards to 1, they DO actually wander between areas within reason, HOWEVER, this only happens when they have nothing to do. If you give them enough items to interact with, they will not get chance due to the day length. The only things they can't do iirc is cross "non-paths", namely, bridges and beaches. Either way, any decently done island will pretty much lock them in one area functionally though yeah.

Keep in mind there is a limit of 2 lodgings in the south side and 3 in the north.
If I place a shop or something to improve guest satisfaction in another area that the NPCs can't physically walk to, will I still get the bonuses or no?

Any of the furniture marked as "UP!" such as dokobucks, popularity, and satisfaction will need the guest to physically interact with said item to make it work. If you purely care about functionality and you don't care for appearance, you want to stack a ton of small ones of these right outside the lodgings.
