Make sure the prestige rewards are maxed, but not the curse level. Biggest gain for least resistance.

Basically, it's using the effect on Contagion that not only consumes all poison stacks, doing the total poison damage all at once, but it also multiplies that damage by a value based on the damage bonuses to that skill itself. (that is, generic damage increases will boost the amount of poison damage per stack, they will not boost Contagions multiplier, but damage bonuses that apply directly to Contagion will increase that multiplier).
So the goal is to apply as many poison stacks as quickly as possible, with as high quality stacks as possible, with as high a multiplier as possible, so that when Contagion procs, it blows everything up.
I recently did a modified version that, where instead of using Devastating Blow to remove 3 skill slots, I used Focused Mind to remove 2, filling it with Festering Strike (adds both poison and weakness to make poison do more). I also changed Dash Mastery and Death Touch for Elemental Flow (for even more poison damage) and Quality Standard (so that every level up gave stronger bonuses). Yes, you only get to chose from 2 instead of 3 with this, and there are more trash upgrades to skip/ignore due to 4 slots instead of 3, but I still had some 40 rerolls at the end of this run, and I was able to clear OL 7 from start to end in under 19 minutes (about 2 minutes faster than Ulyaoth's screenshot in his build guide), with a character who started at prestige level 50 (and thus had a weapon with zero upgrades on it)! ...so it still worked out quite well imo.

Oh, and be sure to turn OFF ascensions, so you don't get ascension related upgrades diluting your upgrade pool as well.
As you can see, that one 19 minute run took me from around 50 to 135.
EDIT: second run only took 16:10 to get the same distance, and got me to 162. One more run will be more than enough to get over 170. So yeah, 3-4 runs at under 20 minutes each is possible for each character.
