1. Stacks confusion debuff on enemies which gives them % chance to miss on every attack. Eventually you will stack hundreds to thousands of stacks of confusion on an enemy almost instantly, this is a must to survive later waves. I don't know what the base % chance to hit for mobs is, but it's impossible to dodge in later waves because enemies attacks instant charge.
2. Has the lasting, empowering, and holy effects and it synergizes really well with the other abilities. After my own experimentation, abilities with "lasting" seem to do the most damage. I've tried runs using Titan's Cleaver and Shield Bash, both abilities perform well at the start, but beacon of light and light beam outstrip them both eventually (plus the debuffs they apply).
3. Eventually has a massive AoE so you debuff/hit enemies even if they're off screen.
I did 13 Endless with it and then just ended my run as loop 13 took ~70 minutes. (Endless is slower due to time scaling hp)
Wp anyway.
First try was with Subdue, to give a larger damage boost but survival became impossible at Loop 9.

So I followed the exact Skill Setup but changed some runes for more surviveability.
I couldnt die since Armor, Block and Confuse combined made taking damage trivial.
But the lack of damage was insane and loop10 alone took ~30 minutes?

More defense was relative unneccessary.
Lost my life due accidently dashing into the Void Presence.
Fiddled around with Runes a bit more, DOTs seem complelty useless, no matter how high they stack at this point. This made me sad, but well so it is.

Still my best run was with Chaoswalker full Chaos Build, although the stats dont match the Skills I used and i cant recall anymore which I used.

I was thinking if barb is the best char for this,...
I might try out a bit more, but i need a break for now.

I ended the runs, because it just took to long and was boring af.
Full defense build could go on way longer definetly.
I stopped at wave 11 because the HP scaling of the enemies was more or less exponential while my damage boosts were linear and very minimal. I tried swapping out the 90% damage versatility rune for the rune that gives you a seventh ability slot but at a -15% damage debuff so I could stack the "Exposed" status trait, but it didn't seem to work any better and was probably a little slower.
TL;DR : IMO anything past wave 10 takes way too long to kill. If there is going to be an endless mode they need to follow in the steps of idle games that will give you universal damage multipliers at certain intervals. Or maybe item pickups that allow you to vary your build even more since you are more or less locked in by level 50 for the rest of your run unless you've had crap luck.

Paladin felt alot better, probably due Disoriented.
Collecting the Ultmate Power was also very manageable, since you can face tank everything except the VP and this gave a very large dmg boost.
Overlord has higher hp scaling and higher damage scaling, endless just has more enemies and less lords
