I dont get this myself.
I have the feeling you have at least one active Skill for each to even get the Synergy Chain Buff, but then I get them sometimes out of the blue.
Sometimes I dont get one offering even with the Synergetic Rune and when I have 6 different Skills, that apply different Buffs.
The behaviour behind this seems very strange.

While the Rune got nerfed and the spreadsheet isnt up to date, it is still a good Rune to pick.
Its not a flat additional Multistrike chance.
There is a general issue with cryptic and sparse informations for those who want to minmax and going deep into numbers.
Some Runes give multiplicative DMG Boni like Generalist, while others give just flat Boni, like Vulnerable Target.
Hope this will get solved in the future.
It has two effects:
Skills that have a synergy (like Bleed causing Poison or Finesse giving Bulwark) have a higher chance to roll (they are rare skills though, so you need to still roll a rare, but if it is a rare it is more likely to be one of them)
Secondly they also increase the chance of these skills triggering. Usually the buff ones have 25% trigger chance and the debuff ones 50%. These are increased to 37.5% and 75% respectively
