It's hard to pick one, but I would definitely say Chapter 1 with the Village fight and Chapter 5 Cabin are pretty good contenders towards being at the top for Professional NG (no bonus weapons), since it's in that weird spot of not having much weaponry at all while still having extremely sturdy enemies, but the cabin fight does get quite a lot easier when you just camp on the 2nd floor as soon as it starts. There is also the chapter with the double Garrador room which can just be super chaotic at times.
I would say that getting the S+ ranks on Pro is gonna rely a lot on Chicago, due to not only being a strong weapon but having Infinite Ammo exclusive, which solves the ammo problem.
My recommendation > immediately flash or grenade them at start when they get to stairs (wait before stairs where you very first start). Run behind them and rifle/riot gun, or if you are going magnum route use that. Kill them in about 2-3 grenade/flash and in like 10-20 seconds.
Cabin is yeah, still a bit of effort.
For me I don't like Ramon for speedrunning pro purposes because the egg's hit detection on him is very finnicky if using that method. The boss also has what I'd consider pretty clear glitches where it skips animation/delays and will instantly charge or shoot in some occasions I've seen in both runs that are physically impossible to dodge. He is suppose to have some animations before his actions but does not always commit them and the issue while not super common occurs at least a few times per fight (1-2 at least) unless you instant kill via gold egg >shoot/stab/rocket methods which isn't solving the issue but outright bypassing its existence.
You don't flash them BEFORE shooting them, you ring the bell, shoot them both in the back, and throw 2 flashbangs, there you go, both dead at the same time
Weirdly, I found that skip actually made it significantly harder.
If you do that then Mike doesn't thin out the huge amount of enemies who will then swarm you. The only way I got through Chapter 15 was to do that bit properly.
If you've been quick in other places then you should have some time to spare by that stage.
I've done it with one grenade, think it can be any, if you go around and behind the building of the turret, and toss it right in on top of the turret.
That stretch is miserable.
For me it was
Village: Canyon, Cabin Fight: Area before Bella Sisters, and Bella Sisters themselves.
Castle: Water Hall, Double Garrador Room, and Salazar.
Island: War Zone with Mike.
I had so many resets because of these areas.
