do i need to purchase every weapon in one playthrough?

No since they carry over in ng+ you can do as many ng+ as you need to get them all

Just buy them all to have the achievement then once you have the achievement resell them all.

Im not sure if you buy and resell instantly will work

That what i did since you can run several ng+ you will have plenty of money for that

You also need all of the unlockable challenge weapons, like the handcannon and thompson.
As far as i tested at least, i didn't get the achievement for owning all regular guns, so special weapons like the mentioned handcannon,thompson SMG and what ever else will be needed too.

Also don't forget about unlockable knives (3 of them).

On the second thought, I guess if it's an unlockable weapon then you still need to obtain it.

Also bear in mind that those unlocked weapons has to go to your case at least once to count as obtained.
