That's exactly why I made this post. I'm currently doing a handgun run only and realized that this is possible. I love this game! :D
You can get the second one with a grenade and completely skip both of those towers
In the castle, shortly before the armored el gigante attacks you, there are some gates with sun and moon symbols and you hit levers to open one and close another. You can skip two of the towers by hitting switches on the other side of gates when you are not supposed to be able to. The first one you can shoot and the second one you can throw a grenade at. Does a regular or heavy grenade work for the mines area? What about the wrecking ball wall scene.
2-3 normal grenades work for the wrecking ball area.
As far as I know you cannot, hence the grenade strategy. Sorry I didn’t clarify (not being sarcastic)
