Why did they want Ashley so bad?

They wanted her to infect her father after being "rescued". I'm guessing it was an easier job to kidnap her then going straight after President Graham himself.

The plan was the kidnap the daughter of the President while she was on spring break or whatever, infect her, force feed her the black water to force the parasite's rapid growth, probably imprint commands on her, then send her back to dear ol daddy and have her infect the President and likely everyone else he comes into contact with, including other world leaders. This would proliferate the complete control of Saddler over every single world's government, but would start with the US.

We all understand that the first medical examination after rescue would uncover whole super duper plot. The poin is Sadler is a ♥♥♥♥♥♥♥ spanish no name clown that lives in some dream world. And most likely parasite inide him influences his mind making him even dumber.

Once the genetically engineered “superior” plaga had fully merged with Ashley they wouldn’t have noticed a difference unless they new what they were looking for, which they don’t. It’s goofy plot, it’s Resident Evil after all, but we’re never given any reason to think Saddler’s plan wouldn’t have worked. Any realistic hole in the plan would just be hand waved away with “because plaga”, which is fine for the B-movie schlock they are going for.

The parasite only damages the brain like that if it's a low grade parasite. Her and Leon were given better variants of the las plagas that keep the host's intelligence in tact and doesn't do the whole head explodey thing. That variant was still below the rank of what Saddler has. Which is why he could control them. Saddler has a more kingly type variant of las plagas. Luis's amber sample also had a more kingly type variant that was as strong as Saddler's variant or even stronger.

We could say it's a more queen-ish type yeah because the whole hive mind stuff and all that. Except Salazar fused with a queen las plagas early in the game, It's all incorrect terminology by me either way. I think the game's lore just says these are a "superior" type or something along those lines.

The whole point is that the OG RE4 Story was written in 2 weeks by Shinji Mikami. They wanted to make an action game - something radically different from previous entries and there is no better excuse to make an action game than 'Hey they captured America's sweetheart want to go invade Spain and blast everyone to kingdom come?'

That's the whole point, the story serves the gameplay purpose; it makes no sense to send in Leon solo with the two most inept Policia Nacional I've ever seen, they just want you to blast your way through Valdelobos without second thought lol, wouldn't be fun if you had a team now would it?

Now in - game lore wise, yeah they Krauser cut a deal with Saddler since his discharge from the Special Forces was due to nerve damage and he was bitter about it so he kidnapped Ashley and they want to control her with the parasite, which makes me wonder how hard that Bluetooth or WiFi or whatever the ♥♥♥♥ Saddler uses to mind control people goes, he must have really good coverage lmfao

not the new version of plaga that they gave Ashley, it's a new better improved version more stable and what not etc etc. It's in one of the notes you can read.
