Master classes are unlocked later in story missions (I believe the first several are unlockable after finishing the 6* main story mission, while the rest are unlockable after the 7* main story mission).
When you change classes, you retain your adventurer's level.
Certain negative traits on unique characters can be removed and replaced with positive traits via certain actions on certain story missions. GreenGuy confirmed just a little bit ago on a different post that the Frail, Dumb, and Deathwish traits have a chance to disappear each day too (after certain conditions are met, you can find the thread titled "Hidden Talent Report" to see his full response there). You can gain new traits via a multitude of events, assignments, during training (again see "Hidden Talent Report"), and just randomly each day (Scarred trait).
thank you
