Personally I like Constantinople more than Rome (or any of the cities in 2); partly because of its look and partly because I felt like it was laid out a little more logically. And I enjoyed ACR more than any of the previous AC games as well. Yes, the story is pretty much just fleshing out the existing plot instead of advancing it, but if you like the story in Assassin's Creed, you'll enjoy Revelation's contribution to it. As for gameplay, if you've played 1, 2 and Brotherhood, then you'll probably get the most out of the fairly minor changes they made in Revelations. The hook-blade improves your climbing a little, the zip lines get you around the city a little faster, and the bombs allow for quick and easy crowd management.
The assassin recruitment/leveling meta game is now larger and serves more of a purpose, and the tower defense mini game they've added doesn't come up very often, but works well as a nice break in the action.
In terms of overall length/content ... I think Revelations was a little shorter than Brotherhood and noticeably shorter than AC2. I bought the game of Amazon and I'm not sure if there's anything in game that tracks play time, but I basically played nothing by ACR for about 5 days until I was done (with about 75% total sync). ACR is a smaller game than 2, but it does everything a little better.
ACB or ACR side quest: here's 4 soldiers that just finished their training and joined the antagonist's army! Go kill them!
AC2 had filler side quests. The other two had side quests that could've been main quests.
