What do you think is the actual relationship between Quote and Curly?

The Iron Bond, the stolen panties, the fact she decides to live together with him, when she blushes at him after regaining her memories... these lead me to believe she likes Quote, but what about him? He's cryptic, to say the least

Curly established long ago that she prefers Quote to stay quiet and out of her way if their relationship is going to work, so that's why he's so "cryptic". And if that sounds jerkish, then, well, it is.

It was a joke anyway. A subtle political joke. So I don't know how else to explain it (unless you want me to get into the actual politics), but I understand that it could be hard to understand, so maybe we should just pretend that I was never here.

Yeah, I'm not good at being serious when trying to figure out stories. But anyway, you know how the mod Jenka's Nightmare claims that they're related? Maybe that's actually canon and Quote knows that, but Curly doesn't, so instead of emoting, he just spends all his time worrying about how he's going to tell her and prepare for her reaction. Or maybe they're not related and he just doesn't like her the way she likes him, which would lead to the same behavior.

I'm trying my best to come up with a serious answer, but maybe I should just leave that to someone else.

Even if they really are related, it wouldn't matter, since they are robots/cyborgs. Futile matters such as "sibling relationships" can't really delete feelings

Well, it can be very complicated, they were both sent to the island 10 years ago and their team purpose was to destroy the demon crown then use it or deliver it then the doctor and the robots, we can get the proof from paying attention to certain dialog during important parts of the game. And when Quote feeds Curly the Pa Migon and restores her/his (depends if your on Story Mode or Curly Mode) you will learn in the dialog. So I think they are best friends or they are a couple.

At the end of the best ending where you see Quote, Curly, and Balarog, flying away, Quote and Curly have a heart appear. I think they love each other and are happy that they saved the Mamiga and humans. Or something like that.

Anything that dies in Cave Story explodes spectacularly, they just hit the floor. Balrog says himself that Misery sent him to rescue Quote and Curly, and for Sue.
