After your first modless playthrough, you will be able to identify what elements of the game annoyed you, and then correct these with mods.
I'm saying this because everyone has different expectations (hence why people play with so many different mods) - and there is no "best" combination of mods. It all comes down to yourself and what you expect from it.
As for Synergies, you won't really appreciate the mod unless you play the vanilla version of TL2 first. Instead of being like "wow it changes so much the game", you will be more like "yeah it's a good ARPG".
The above statements are true.
However some players may wish to eat the cake and skip the salad... ^_^
I will provide a list of links to Synergies friendly mods below:

One of the best modded endless dungeon challenges.

There are other mods but I can confirm that the above list works with Synergies.

I would recommend other mods besides these but I have yet to confirm that they work with Synergies.

zoomy allows you you to zoom the camera much further out than normal, and as a side effect, it fixes the bug above. It is definately compatible with Synergies. I would NOT play TL2 without this mod!!
