Offense and defense affects clash power, every 5 points difference adds a +1 final clash power to the one with higher offense/defense.

1-2-3 are uptie tiers. If you haven't completed chapter 2 yet, I'm not sure if you can actually use that feature? Been a while, anyway. Once you get there, you can use Thread (From daily luxcavation, battlepass, converting spare Egoshards, etc) to "Uptie" an Identity (or E.G.O) which will boost it, as Silver described.
The first Uptie boosts their first and second skills, unlocks their "in the team" passive ability, and increases their speed (So a Sinner who's at, to pull a number at random, at speed 3-5 will every turn have speed 3, 4 or 5. Uptied, they'd have speed 4, 5 or 6.)
The second Uptie unlocks their third skill (-usually- the most powerful), provides a boost to the numbers on their first two skills (either base value - what you always get - or coin value - what you get added to base value if you flip heads) and another +1 to speed.
