It does not. In general, any items found on the floor laying randomly don't respawn. There are locations that respawn clay if you progress past the island of hope

Clay does respawn at the ground level mine pits just not the open to pick up spots by water collectors

There is also the termite mounds that give clay and respawn on Copper Island in the 2nd region.
