How do upgrades work?
Can you take both paths in a tree, or does taking one lock you out of the other? Do you need BOTH paths to unlock the final upgrade?

You can take a path in both trees.
Unlocking one talent locks you into that talent, till you get a second one of the same tier in the same tree to switch between
No, they tiers are unlocked based on total skills levels. you can max all 3 Tier 1 skills (15 points total) to reach Tier 4

The other person who commented is talking about the rune upgrades in the character select menu. For the in-game upgrades, taking one path does not lock you out of the other. And you only need one of the two middle upgrades to be able to roll the final upgrade. You can still roll the other middle upgrade after you have the final one. It's very forgiving in that sense :)
