Do you always try to clear every lair boss? If so what team do you find that can consistently clear Tangle or Sprawl as the first region without losses?
Not always, I do not care about lairs if I'm farming candles. Clearing all lairs do give a hefty candle bonus, but it takes a bit too much time and it is somewhat risky and I often have to neglect the region goal in order to clear a lair. Clearing 2 or less lairs is not worth it just for the candles. If I want a chance against the confession boss, clearing all lairs can help immensely with trinkets, relics and baubles tho.
Frankly I choose heroes based on their quirks and I do not try to decide my tactics right at the start, trinkets, aquired quirks, even stagecoach items and combat items can have so much impact it would be really hard to force a specific tactic throughout the whole run so I just work with what I get. So far it seems to me that almost any combination of heroes is viable as long as you have the unlocks for them, but sometimes I do feel sprawl/tangle boss would be too risky for my current team as a first lair boss and I try another region.
This is one of the smartest pieces of advice- you don't realize until many run throughs that because the 'stuff' you get each run is so random you are better off to adapt to what you have than stick to something you want to do. The game is always going to give you lemons, whether you can turn that into lemonade or not is the game
1) Once team is made I already know which lair boss I want to take out. I will try to avoid going to that region right away and save it if possible for second or third.
2) The most important that basically overrules everything, what the region gives. I know you said no factors but that just what I do. See what I need most and see if the condition is favorable and then go there.
However lets say there is a case where the conditions are not what I like on both options and lair boss already been slain, then I will likely go to sprawl or tangle, both are not too hard if you know what to avoid.

But yeah, remember lair bosses. Most teams can fight leviathan with some set up, a lot can do librarian with some set up, and general needs the right team/set up
Whichever area has what I guess will have the easiest lair boss (normally that's the librarian).
