You then press "Q" , which in turn brings up a "Call Poirot" dialog box at the bottom of the screen (or alternatively "Call Mrs Locke").
And indeed, you have to switch back and forth between the two characters a number of times, as they help each other in their search (one discovers clues/codes that the other can use on his/her side, etc.).
The ability to switch between Locke and Poirot should be unlocked after glancing at everything in the office.
The combination for the Tower of Babel is found in Wadi's apartment. You're not supposed to open it before switching to Poirot once. If you've somehow guessed or brute-forced the combination you might have skipped ahead and soft-locked the game. In that case restarting the chapter might help.
