The difficulty in each run of Gauntlet Mode can pretty rapidly increase, and you'll eventually be getting one-shot. It's VERY HEAVILY recommended that you get a good team of 5 characters you enjoy playing leveled up and geared with some good weapons with good effects via Story Mode before you tackle Gauntlet Mode.
Levels carry on but I don't think you can unlock characters in Gauntlet unless you got them from the story mode, I may be wrong.
Correct! Gauntlet can indeed be used for leveling, though it's risky if you don't make sure you're ready to exit at a moment's notice - and also correct, unlocking characters in Gauntlet does not unlock them in Story!
Also in the first few chapters in the story, what characters can you recommend?
I don't recommend any particular characters because these games aren't designed for there to be a super obvious best who you should always have on your team, etc. The series has, for a very long time, been all about picking your favorite and going to town on enemies with them.
By extension of how mechanics work in later stuff, some characters perform better than others, but you're gonna be able to tear through enemies with basically anyone after you level them up and get upgraded weapons, so try out as many as interest you, and keep leveling the ones you end up loving the playstyle of!
