The final boss of that game is called "Etreant". Which might be a portmanteau of the town's name and the ancient tree monster. So, "Etrian" is just an adjective for "monster tree". Although, I did some etymological digging, and it turns out that the root of THAT word is just "giant" ("ent"). So, I don't really know, but, at the same time, I don't think it's just the name of a town. What is the town's name in Japanese? That might give some more clues, especially considering that Japanese is spelled as a phonetic language... there's no difference between "Etria" and "Etrea" there.
I can't find anything clear sadly.

and even there, trivial sections, is says:
Etreant's name is likely a play on the words Etrian and Treant, which is a fictional walking tree originating from Dungeons & Dragons.
